How Are Y-STR DNA Test Results Interpreted?

Y-STR results must be interpreted as evidence about a paternal male lineage, rather than as a simple identification of one specific family relationship.

Profiles Consistent If the tested males have matching or appropriately consistent Y-STR profiles, the DNA evidence can support that they share a common paternal male lineage.
Profiles Not Consistent Significant incompatibility between the Y-STR profiles can exclude the proposed shared paternal lineage between the tested males.
A matching Y-STR profile does not identify the exact relationship. Men belonging to the same paternal line may have very similar Y-STR profiles. Therefore, Y-STR testing alone generally cannot distinguish whether two matching males are father and son, brothers, paternal uncle and nephew, grandfather and grandson, or another relationship along that same male line.

Who Can Take a Y-STR Relationship Test?

Direct Y-STR testing requires participants who have a Y chromosome, because the test analyses markers located on the Y chromosome and follows father-to-son inheritance.

Possible participant combinations can include appropriately related males such as:

Two alleged brothers
Male child and alleged paternal uncle
Male child and alleged paternal grandfather
Male cousins connected through the paternal male line
Males from different generations of the same proposed paternal line
Other males connected through an uninterrupted father-to-son lineage

Can a Woman Use Y-STR Testing to Investigate Her Paternal Family?

A woman cannot provide a Y-chromosome sample for direct Y-STR analysis because she does not have a Y chromosome.
